ELISA Kit for the Detection of Native and Recombinant Protein A

Protein A capture step (Non-MabSelect SuRe ™, PrismA or NGL)를 활용하는 downstream 공정을 위해, native 및 재조합 protein A 리간드 변이체를 포함한 잔여 Protein A를 정확하고 정밀하게 정량화하도록 설계되었으며, 이 키트는 단일클론 항체 정제를 위한 친화성 크로마토그래피에 사용되는 여러 Protein A 리간드 중 하나인 Repligen의 재조합 Protein A standard (rProtein A™)가 함께 제공됩니다.

Repligen의 Protein A ELISA 키트는 적절한 절차에 따라 천연 황색포도상구균 Protein A 및 기타 재조합 Protein A 형태를 포함한 다른 Protein A 변종을 정량화하는 데에도 사용할 수 있습니다.

Protein A ELISA kit는 치료용 단백질 제품에 존재하는 소량의 Protein A를 측정하는 신뢰할 수 있고 사용하기 쉬운 고감도의 분석법으로 다음과 같은 여러 개발 및 상업용 제조 단계에서 사용 가능합니다

  • 공정 개발: 특정 조건에서의 resin 침출 (leaching) 특성
  • 제조: 정제 공정의 여러 지점에서 채취한 침출 샘플
  • 완제품 출시: 공정 억제 수준 및 lot간 일관성 문서화

  • Sensitivity: 0.1 ng / mL or 0.8 ppm
  • Recovery: 80-120%
  • Accuracy and precision: Inter-assay +/- 10%, Intra-assay +/- 15%

Kit Contents:

  • Anti-protein A coated 96-well microtiter plate

  • Reagent A: sample diluent (5x)

  • Reagent B: recombinant protein A standard solution

  • Reagent C: rabbit anti-protein A: biotin probe

  • Reagent D: streptavidin: peroxidase conjugate

  • Reagent E: TMB peroxidase substrate

  • PBS packs (quantity 2)

 

How To Use:

  • 이 키트에 제공된 polystyrene microtiter plate는 anti-Protein A antibodies로 코팅되어 있습니다.

  • rProtein A™ standard (Reagent B) 및 테스트 샘플은 샘플 희석액(Reagent A)과 함께 희석하여 immobilized 항체와 함께 인큐베이션합나다.

  • Captured rProtein A™ 리간드는 이후 Biotinylated anti-Protein A probe (Reagent C)를 추가하여 검출합니다. 이 probe는 높은 치환율을 가지고 있어 Streptavidin Peroxidase conjugate (Reagent D)의 최대 결합을 가능하게 합니다.

  • 최종 검출 단계에서는 Tetramethylbenzidine, TMB (Reagent E)를 추가하여 매우 민감한 발색 반응을 일으킵니다. 발색 강도는 샘플에 존재하는 rProtein A™ 리간드 양에 비례합니다.
